How Do I Create a Flight Route?(I read the wiki, but don't know how to actually establish a route.)

Hi everyone who's reading this. I have an airport in my city of Paskua, on the island of Lorotoban. I have seen the wiki for how to find destination airports, but don't know how to actually establish a route. How would I do this?
